چکیده

In cellular, tissue-level or whole organ bioreactors, the level of dissolved oxygen is one of the most important factors requiring control. Hypoxic environments may lead to cellular apoptosis, while hyperoxic environments may lead to cellular damage or dedifferentiation, both resulting in loss of overall tissue function. This manuscript describes the creation, characterization and validation of a bioreactor system that can control oxygen delivery based on real-time metabolic demand of cultured whole lung tissue. A mathematical model describing and predicting gas exchange within the tunable bioreactor system is developed. In addition, the inherent gas exchange properties of the bioreactor and the inherent oxygen consumption rates of native rat lungs are determined, thereby providing a quantitative relationship between system parameters and levels of dissolved oxygen. Finally, the mathematical model is validated during whole lung culture under a range of system parameters. The system presented here provides a quantitative relationship between the concentration of dissolved oxygen, tissue oxygen consumption rates, and controllable system parameters that introduce gasses into the bioreactor. This relationship not only enables the maintenance of constant levels of dissolved oxygen throughout a culture period during which cells are replicating, but also provides noninvasive and real-time estimation of the metabolic and proliferative states of native or engineered lung tissue simply through dissolved oxygen measurements. Lung bioengineering: physical stimuli and stem/progenitor cell biology interplay towards biofabricating a functional organ

A current approach to obtain bioengineered lungs as a future alternative for transplantation is based on seeding stem cells on decellularized lung scaffolds. A fundamental question to be solved in this approach is how to drive stem cell differentiation onto the different lung cell phenotypes. Whereas the use of soluble factors as agents to modulate the fate of stem cells was established from an...

Performance of Biological hydrogen Production Process from Synthesis Gas, Mass Transfer in Batch and Continuous Bioreactors

Biological hydrogen production by anaerobic bacterium, Rhodospirillum rubrum was studied in batch and continuous bioreactors using synthesis gas (CO) as substrate. The systems were operated at ambient temperature and pressure. Correlations available in the literature were used to estimate the gas-liquid mass transfer coefficients (KLa) in batch reactor. Based on experimental results for the con...

Pulmonary gas-exchange analysis by using simultaneous deposition of aerosolized and injected microspheres.

Numerical methods for determining end-capillary gas contents for ventilation-to-perfusion ratios were first developed in the late 1960s. In the 1970s these methods were applied to validate distributions of ventilation-to-perfusion ratios measured by the multiple inert-gas-elimination technique. We combined numerical gas analysis and fluorescent-microsphere measurements of ventilation and perfus...

Mixing Studies in Loop Bioreactors for Production of Biomass from Natural Gas

The mixing behavior of the gas-liquid phase in three loop bioreactors was investigated. A gas-induced External Airlift Loop Bioreactor (EALB), a forced-liquid Vertical Tubular Loop Bioreactor (VTLB) and a forced-liquid Horizontal Tubular Loop Bioreactor (HTLB) were used for mixing studies as well as biomass production from natural gas. The effect of design parameters, riser to downcomer cro...

Physiological evaluation of a new quantitative SPECT method measuring regional ventilation and perfusion.

We have developed a new quantitative single-photon-emission computed tomography (SPECT) method that uses (113m)In-labeled albumin macroaggregates and Technegas ((99m)Tc) to estimate the distributions of regional ventilation and perfusion for the whole lung. The multiple inert-gas elimination technique (MIGET) and whole lung respiratory gas exchange were used as physiological evaluations of the ...
